白香丹胶囊对PMS肝气逆证模型大鼠海马GABA_A R β_2亚基蛋白分布模式及表达水平的影响

摘要(Abstract):

目的:探讨中药复方白香丹胶囊(BXD)对经前期综合征(premenstrual syndrome,PMS)肝气逆证模型大鼠海马中γ-氨基丁酸A受体(GABAAR)β2亚基蛋白分布模式及表达水平的影响。方法:阴道涂片镜检法和旷场实验筛选合适大鼠,随机分为正常组、肝逆组和肝逆给药组(BXD给药剂量10 g.kg-1.d-1,给药5 d);电刺激法复制PMS肝气逆证模型,同时采用宏观行为学观察和旷场实验来评价模型;免疫荧光技术和蛋白印迹技术(Western Blot)测定大鼠海马神经元中GABAA R β2亚基蛋白分布模式及表达水平。结果:与正常组相比,肝逆组大鼠旷场实验得分明显增加,海马神经元GABAAR较集中分布于组织边缘,β2亚基蛋白表达水平显著升高(P<0.01);而造模同时给予BXD后,大鼠旷场实验得分明显减少,海马神经元GABAA R分布模式无明显变化,而β2亚基蛋白表达水平显著降低(P<0.05)。结论:大鼠海马神经元GABAA R β2亚基蛋白表达水平上调可能是与PMS肝气逆证密切相关的重要微观机制之一;中药复方BXD可对GABAA R β2亚基蛋白表达水平变化起到纠正作用。
Objective:To explore the effects of Baixiangdan capsule(BXD),a Chinese herbal compound,on the location and expression levels of GABAA receptor β2 subunit(GABAARβ2) in hippocampus of PMS model rats with liver-qi invasion.Method:After vaginal smear examination and open field test,the selected SD rats were randomly divided into 3 groups: Normal group,PMS model group with liver-qi invasion and PMS BXD-administration group with liver-qi invasion(BXD was administered with oral dosage of 10 g·kg-1 body weight every day for 5 days).PMS model rats with liver-qi invasion were induced by electric stimulating,and evaluated by macro-behavior observation and open-field test.The location and expression of GABAA receptor in hippocampus were measured by fluorescence microscopy and western blot respectively.Result:Compared with the normal group,the open field scores and GABAARβ2 expression of PMS model rats with liver-qi invasion were increased significantly,and the distribution of GABAA receptor is more concentrated.However,the scores and GABAARβ2 expression of PMS BXD-administration group with liver-qi invasion were decreased markedly.Compared with the PMS model group the location had no significant change.Conclusion: One of micro-mechanisms of PMS model rats with liver-qi invasion may be related with the high expression of GABAA Receptor β2 subunit in hippocampus,and the Chinese medicinal formula,BXD granule,had an adjust on the above abnormal changes.
