水翁花对微粒体和神经细胞氧化损伤的保护作用

摘要(Abstract):

目的 :研究水翁花对小鼠肝微粒体膜脂氧化及H2 O2 诱导的PC12神经细胞氧化损伤的保护作用。方法 :用小鼠肝微粒体膜脂氧化模型、细胞培养MTT法测定细胞的存活率等方法。结果 :水翁花水提物能强烈抑制小鼠肝微粒体膜脂氧化 ,IC50 为 0 .0 13g·L-1,效果优于阳性对照维他命E(IC50 为 0 .0 6 4 g·L-1) ;对H2 O2 引起的PC12神经细胞氧化损伤亦显示出较强的抑制作用 ,对神经细胞表现出保护作用。结论 :水翁花对膜脂氧化及神经细胞的氧化损伤的保护作用 ,可能用于治疗与氧化损伤有关的脑疾。
Objective: To observe the protective effects of Cleistocalyx operculatus on lipid peroxidation in rat liver microsomes and on the trauma of PC12 cells induced by H 2O 2. Method: The mouse liver homogenate lipid peroxidation assay and PC12 Cell culture and Cell viability (MTT assay) were applied. Result: Cleistocalyx operculatus showed strong protective effects on lipid peroxidation in rat liver microsomes in a dose-dependent manner and exhibited potent protective effects on the trauma of PC12 cells induced by H 2O 2 (200 μmol·L -1 ) when the concentration reached 1.00 g·L -1 . Conclusion: Cleistocalyx operculatus may be used as antioxidant to prevent or delay the pathogenesis of neural cell diseases.
