反相HPLC同时测定白花蛇舌草口服液中4种组分含量

摘要(Abstract):

目的:建立反相HPLC同时测定白花蛇舌草口服液中3,4-二羟基苯甲酸甲酯(Ⅰ)、对香豆酸(Ⅱ)、阿魏酸(Ⅲ)和反式6-O-对香豆酰鸡屎藤苷甲酯(Ⅳ)的含量。方法:采用DiamonsilTMC18柱(4.6 mm×250 mm,5μm);流动相A乙腈,B甲醇-水-冰醋酸(5∶95∶0.25),梯度洗脱:0~20 min,1%~16%A;20~42 min,16%A;42~46min,16%~20%A;46~65 min,20%A。检测波长265 nm;流速1.0 mL.min-1。结果:Ⅰ,Ⅱ,Ⅲ与Ⅳ分别在2.1~105(r=0.9998),3.5~175(r=0.9998),1.72~86(r=0.9999),4.0~200 mg.L-1(r=1.0000)线性关系良好,方法平均回收率分别为99.9%,97.9%,98.6%,98.1%。结论:方法简便快速、准确度高,可用于白花蛇舌草口服液中4种组分的同时测定。
Objective: A new method for simultaneous determination of four methyl-3,4-dihydroxybenzoate(Ⅰ),P-coumaric acid(Ⅱ),ferulaic acid(Ⅲ) and E-6-O-P-coumaroyl scandoside methyl ester(Ⅳ) in Hedyotis diffusa oral solution by reversed-phase HPLC was developed.Method: The separation was performed on a DiamonsilTM C18 column(4.6 mm×250 mm,5 μm) with gradient elution.A-acetonitrile,B-methonal-water-glacial acetic acid(5∶95∶0.25),0-20 min,1%-16%A;20-42 min,16%A;42-46 min,16%-20%A;46-65 min,20%A.The UV detection was set at 265 nm;flow rate 1.0 mL·min-1.Result: There was good linearity between the peak area and concentration at the ranges of 2.1-105(r=0.999 8),3.5-175(r=0.999 8),1.72-86(r=0.999 9),4.0-200 mg·L-1(r=1.000 0) for Ⅰ,Ⅱ,Ⅲ and Ⅳ respectively.The average recoveries of Ⅰ,Ⅱ,Ⅲ and Ⅳ were 99.9%,97.9%,98.6% and 98.1%.Conclusion: The method is rapid,simple and accurate,and it can be used for the evaluation of H.diffusa oral solution.
