白芍中芍药苷及其衍生物的UPLC-MS/MS分析

摘要(Abstract):

目的:采用超高效液相色谱与四级杆-线性离子阱串联质谱联用技术(UPLC-Q-Trap MS-MS)对白芍中单萜糖苷类成分进行分析和鉴别。方法:用Acquity UPLC BEH C18色谱柱,以0.1%甲酸水溶液(A)乙腈(B)为流动相梯度洗脱,使用ESI离子源,在负离子模式下采集数据,数据采集范围m/z100~1200,雾化气124.11 kPa,脱溶剂气103.43 kPa,气帘气206.85kPa;离子源温度为450℃;离子喷雾电压为-4.5 kV;结果:白芍中化学成分获得了较好的分离和检测,共推测出6个糖苷类化合物的化学结构;结论:该方法灵敏度高,分离度好,适用于白芍中单萜糖苷类成分的快速分析与结构鉴定。
Objective: To investigate the monoterpene glycosides in Paeonia lactiflora by UPLC-MS/MS.Method: An Acquity UPLC BEH C18 column(2.1 mm × 50 mm) with 1.7 μm particle size was used.The mobile phase was composed of acetonitrile and 0.1% formic acid in gradient mode.The flow rate was 0.4 mL·min and the chromatographic run time was 9 min for one run.The mass spectrometer equipped with an eletrospray ion source in negative ion mode.Results: Totally six glycosides were analyzed and identified by the established UPLC-MS/MS method.Conclusion: The method was rapid,sensitive,and extremely useful for rapid identification of glycosides in P.lactiflora.
