甘草制吴茱萸炮制研究

摘要(Abstract):

目的:通过正交设计建立甘草制吴茱萸的炮制工艺,并进行质量标准研究、毒理研究和药效学研究。方法:以吴茱萸胺、吴茱萸次碱为指标进行炮制工艺的优化,以RP-HPLC测定吴茱萸胺和吴茱萸次碱的含量,并进行吴茱萸生品和甘草制品小鼠的急性毒性实验和药效学试验。结果:甘草制吴茱萸的炮制工艺,甘草汁用量以6倍于药材的甘草溶液进行闷润,以低于230℃炒制至干。毒性及药效研究:吴茱萸生品和甘草制品以40(g饮片).(kg体重)-1,b.w给小鼠一次性灌胃给药后观察7 d,各给药组的动物均未见明显异常;给小鼠灌胃给予0.6 g生药.kg-1(临床等效剂量)的吴茱萸饮片产品(生品、甘草制品)后,均可明显升高小鼠热板痛阈值;明显减少由冰醋酸诱导的小鼠腹膜刺激性扭体反应次数,对小鼠扭体反应次数,对小鼠扭体反应的潜伏期也有一定延长作用。结论:生品吴茱萸和甘草制吴茱萸的毒性较低,甘草制吴茱萸具有镇痛作用。
Objective:To establish the processing method of fructus evodiae and its standard for quality control,toxicity aspects and pharmacodynamics were carried out at the same time.Method:In the studies of processing techniques,the optimized technical parameters were determined by the contents of evodiamine and evodine.And the acute toxicity and pharmacodynamics were studied by rats.Result:The process was that the liquorice-processed fructus evodiae was wetted by liquorice decoction by sixth of raw fructus evodiae(V/W) and fried below 230 ℃.The method of detecting the contents of evodiamine and evodine was that Alltima ODS C18(4.6 mm×250 mm,5 μm);mobile phase acetonitrile-water-tetrahydrofuran-phosphoric acid(51∶48∶1∶0.05);column temperature 25 ℃;mobile rate 0.8 mL·min-1;wave length 225 nm.The toxicity experimentation show that rats didn't show any notable changes after affused the raw material and the processed fructus evodiae's decotion 40 g·kg-1 b.w.at one time seven days constantly.The analgesic effect was observed after 0.6 g(material)·kg-1(weight) b.w.Conclusion:The toxicity of the raw material and the processed one were low and the liquorice-processed fructus evodia analgesic effect was good.
