高效液相色谱法同时测定款冬花中芦丁、异槲皮苷、绿原酸的含量

摘要(Abstract):

目的:建立同时测定款冬花中芦丁、异槲皮苷、绿原酸含量的高效液相色谱分析方法。方法:高效液相色谱法,色谱柱Phenomex Synergi POLAR-RP 80A(4.6 mm×250 mm,4μm);流动相A相为甲醇,B相为乙腈,C相为0.2%甲酸溶液,线性梯度洗脱;检测波长255 nm;流速1.2 mL.min-1。结果:芦丁和异槲皮苷在0.2~2 000μg.L-1、绿原酸在10~2 000μg.L-1与峰面积呈良好的线性关系;平均加样回收率分别为99.5%,100.1%,99.4%,RSD分别为1.0%,2.5%,2.7%。结论:该法简便、重复性好、各成分分离良好,可用于款冬花的质量评价。
Objective:To develop an HPLC method for simultaneous determination of rutin,isoquercitrin and chlorogenic acid in Farfarae Flos.Method: The analysis was carried out on a Phenomenex Synergi POLAR-RP 80A column(4.6 mm×250 mm,4 μm) with gradient elution using methanol-acetonitrile-water(adjusted to pH 2.5 with formic acid) as mobile phase.The flow rate was 1.2 mL·min-1 and the detection wavelength was at 255 nm.Result: The calibration curves were linear over the range of 0.2-2 000 μg·L-1 for rutin and isoquercitrin,10-2 000 μg·L-1 for chlorogenic acid,respectively.The average recoveries were 99.5% for rutin,100.1% for isoquercitrin and 99.4% for chlorogenic acid,respectively,with RSD not more than 3.0%.Conclusion: The described method is reliable and could be used for the quality control of Farfarae Flos.
