吴茱萸及其习用品药材中7个成分的HPLC含量测定

摘要(Abstract):

建立HPLC同时测定吴茱萸及其习用品药材中柠檬苦素、吲哚类生物碱(14N-甲酰二氢吴茱萸次碱、吴茱萸碱和吴茱萸次碱)及喹诺酮类生物碱(1-甲基-2-正十一烷基-4(1H)-喹诺酮、吴茱萸卡品碱和二氢吴茱萸卡品碱)含量的方法。采用YMC C18(4.6 mm×250 mm,5μm)色谱柱;流动相为乙腈(A)-四氢呋喃(B)-5 mmol·L-1醋酸铵缓冲液(pH 3.8)(C),梯度洗脱;流速1.0 mL·min-1;柱温30℃;检测波长220,250 nm。7个成分的分离度良好,标准曲线在检测范围内均呈良好线性,r=0.999 9。加样回收率平均值为96.69%~102.4%,RSD为1.4%~3.1%。该方法简便、准确,重复性好。对5种18批吴茱萸类药材进行含量测定,结果显示7个被测成分在5种药材中均有分布,但含量差异很大。16批正品吴茱萸中7个成分的总质量分数在9.46~69.9 mg·g-1,平均为28.2 mg·g-1。密果吴萸中7个成分的总含量为25.8 mg·g-1,在正品含量范围内,略低于平均值;臭辣吴萸中7个成分的总质量分数为7.69 mg·g-1,低于正品的含量下限。密果吴萸和臭辣吴萸与正品吴茱萸相比,主要成分一致,含量总体低于正品吴茱萸。
This study is to develop a HPLC method for quality evaluation of Euodiae Fructus and related species by simultaneous determination limonin,indole alkaloids( 14-fomyldihydroxyrutaecarpine,evodiamine,rutaecarpine),and quinolone alkaloids[1-methyl-2-undecyl-4( 1H)-quinolone,evocarpine,dihydroevocarpine]in the fruits of five Evodia species. Samples were analyzed on a YMC C18column( 4. 6 mm × 250 mm,5 μm) eluted with mobile phases of acetonitrile( A),tetrahydrofuran( B),and a buffer solution of 5 mmol·L-1ammonium acetate( pH 3. 8)( C) in a linear gradient mode. The column temperature was 30 ℃ and the flow rate was 1. 0 mL·min-1. The PDA detector wavelengths were set at 220 and 250 nm. The seven compounds were well separated and showed good linearity( r = 0. 999 9) within the concentration ranges tested. The mean recoveries were between 96. 7%-102. 4%( RSD1. 4%-3. 1%). Through the validation,the method was proved to be accurate and repeatable. All the seven constituents were detected in the fruits of five species,but the contents of them varied widely in different samples. The total contents of seven constituents in 16batches of Euodiae Fructus were 9. 46-69. 9 mg·g-1,and the mean content was 28.2 mg·g-1. The total content of seven constituents in E. compacta and E. fargesii was 25. 8,7. 69 mg·g-1,respectively.
