HPLC测定菝葜茋类提取物在大鼠体内的分布

摘要(Abstract):

目的:研究菝葜茋类提取物在大鼠体内的组织分布规律。方法:菝葜茋类提取物大鼠灌胃给药1 g.kg-1后,应用HPLC测定氧化白藜芦醇和白藜芦醇在大鼠心、肝、脾、肺、肾中的含量。Zorbax SB-C18色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈(A)-水(B)进行梯度洗脱;流速1.0 mL.min-1;检测波长320 nm;进样量10μL;柱温40℃。结果:建立了氧化白藜芦醇和白藜芦醇在大鼠心、肝、脾、肺、肾中的含量测定方法。结论:本方法简便、准确、专属性强,可以成功应用于大鼠灌胃给药菝葜茋类提取物后,氧化白藜芦醇和白藜芦醇在大鼠体内组织分布的研究。
Objective: To investigate the distribution in rat after oral administration of Smilax china extract.Method: Oxyresveratrol and resveratrol were quantitatively determined by HPLC/DAD method in rat tissues including heart,liver,spleen,lung and kidney after oral administration of S.china extract.The separation was performed on a Zorbax SB-C18 column(4.6 mm×250 mm,5 μm) with acetonitrile-water using linear gradient elution and UV detection at wavelength of 320 nm.The flow rate was 1.0 mL·min-1 and the column was kept at 40 ℃.Result: The HPLC/DAD method for the simultaneous determination of oxyresveratrol and resveratrol in rat tissues was developed.Conclusion: The method has been successfully applied for the distribution study of two active constituents in rat through oral administration of S.china extract.
