高效液相色谱法同时测定复方当归栓中5种活性组分的含量

摘要(Abstract):

目的:建立一种高效液相色谱分析方法,用于同时测定复方当归栓中5种活性成分苦参碱、氧化苦参碱、阿魏酸、左旋紫草素和β,β-二甲基丙烯酰阿卡宁的含量。方法:采用HPLC法,Kromasil ODS色谱柱(4.6 mm×250 mm,5μm),甲醇-0.1%三乙胺水溶液(用磷酸调节pH3)梯度洗脱,分别在检测波长220,316,516 nm,测定了苦参碱和氧化苦参碱、阿魏酸、左旋紫草素和β,β-二甲基丙烯酰阿卡宁的含量。结果:通过1次进样,同时测定了复方当归栓中理化性质差异较大的5种活性成分的含量,且线性关系良好(r>0.999 6),平均回收率为96.92%~102.22%,RSD<3.1%。结论:该法简便、灵敏、准确、重复性好,可用于同时测定该制剂中5种活性成分的含量,并为该制剂质量控制提供了方法学基础。
Objective: To develop a HPLC method for determination of matrine,oxymatrine,ferulic acid,L-shikonin and β,β-dimethylacrylshikonin in compound preparation of Dangguishuan.Method: The chromatographic separation was performed on a Kromasil ODS C18 column(4.6 mm×250 mm,5 μm) maintaining at 30 ℃ during the whole process.The mobile phase consisted of methanol and 0.1% triethylamine aqueous solution(adjusted with phosphoric acid,pH 3)at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 220 nm for matrine and oxymatrine,316 nm for ferulic acid,516 nm for L-shikonin and β,β-dimethylacrylshikonin,respectively.Result: All the compounds showed good linearity(r>0.999 6)in the range of the test concentrations,and the average recoveries of the method is between 96.92% and 102.22%,RSD<3.1%.Conclusion: The method is proved to be credible,sensitive,accurate and repeatable.It can be applied to determine of matrine,oxymatrine,ferulic acid,L-shikonin and β,β-dimethylacrylshikonin in compound preparation of Dangguishuan simultaneously,and provide a basal method of quality control to this preparation and other relative preparations.
