高效液相色谱法测定夏天无中4种生物碱的含量
Determination of four alkaloids in Corydalis decumbens by HPLC

摘要(Abstract):

目的:建立同时分离测定夏天无中原阿片碱、盐酸巴马亭、比枯枯灵、四氢巴马亭4种生物碱含量的高效液相色谱方法。方法:采用ZORBAX Eclipse-C18色谱柱(4.6 mm×150 mm,5μm),流动相为20 mmol.L-1乙酸铵-乙腈,梯度洗脱,流速为1.0 mL.min-1,检测波长为280 nm,柱温为30℃,进样量为10μL。结果:在本实验条件下,4种生物碱有很好的分离,原阿片碱、盐酸巴马亭、比枯枯灵、四氢巴马亭的质量浓度与峰面积分别在1.44~46.0(r=0.999 4),1.26~40.2(r=0.999 8),1.37~44.0(r=0.999 9),1.36~43.6 mg.L-1(r=0.999 9)呈良好的线性关系,平均回收率依次为98.2%,101.9%,102.8%,98.8%,RSD为2.7%,2.5%,3.5%,3.1%。结论:该方法简便易行、准确、重复性好,可用于夏天无中原阿片碱、盐酸巴马亭、比枯枯灵、四氢巴马亭4种生物碱含量测定。
Objective: To establish a quantitative HPLC method for determination of protopine,palmatine hydrochloride,bicuculline and tetrahydropalmatine,in Corydalis decumbens.Method: The separation was performed on a ZORBAX Eclipse XDB-C18 column(4.6 mm×150 mm,5 μm) at a flow rate of 1.0 mL·min-1 using mixtures of two solvents [A(20 mmol·L-1 ammonium acetate)-B(acetonitrile)]: with a gradient elution.The column oven temperature was 30 ℃ and the detection wavelength was set at 280 nm.Result: The 4 alkaloids were well separated by this HPLC method.Linearifies of protopine,palmatine hydrochloride,bicuculline and tetrahydropalmatine were good in the ranges of 1.44-46.0(r=0.999 4),1.26-40.2(r=0.999 8),1.37-44.0(r=0.999 9),and 1.36-43.6 mg· L-1(r=0.999 9),respectively.The average recoveries were 98.2% with RSD 2.7% for protopine,101.9% with RSD 2.5% for palmatine hydrochloride,102.8% with RSD 3.5% for tetrahydropalmatine,and 98.8% with RSD 3.1% for tetrahydropalmatine.Conclusion: This method is proved to be convenient,reliable and accurate.,and it can be used for quality control of C.decumbens.
