摘要(Abstract):
目的:建立同时测定黄连黄柏中木兰花碱含量的液相色谱-质谱联用法。方法:样品经甲醇提取后,采用AgilentEclipse XDB-C18ODS色谱柱(4.6 mm×150 mm,5μm)分离,流动相甲醇-水(60∶40);流速0.8 mL.min-1。质谱采用电喷雾离子化(ESI)方式,正离子采集,以多离子反应监测(MRM)模式对木兰花碱进行含量测定。结果:木兰花碱在4.3522 720pg.L-1线性关系良好。黄连、黄柏平均加样回收率均在98%以上。结论:该方法简便、灵敏、准确,可用于黄连黄柏中木兰花碱的含量测定,为该药材的质量控制提供依据。
A rapid and specific high performance liquid chromatography-mass spectrometric method was developed for determination of magnoflorine in Rhizoma Coptidis and Cortex Phellodendri Chinensis.Samples were extracted by methanol.Agilent Eclipse XDB-C18 ODS column(4.6 mm×150 mm,5μm)was used,and mobile phase was methanol-water(60∶40)at a flow rate of 0.8 mL·min-1.Electrospray ionization model(ESI),and MRM model were used for quantification.The linear range of magnoflorine was 4.352-2 720 μg·L-1.The average recovery was above 98%.The method is simple,sensitive and accurate,it can be used for determination of magnoflorine in Rhizoma Coptidis and Cortex Phellodendri Chinensis.
