中文摘要:该文尝试建立麝香体外抗凝血酶活性效价测定方法,探索品种和产地对麝香抗凝血酶生物活性的影响,为麝香质量生物评价提供科学依据。采用凝血酶滴定法,考察麝香供试品溶液浓度、纤维蛋白原浓度、凝血酶浓度、凝血酶滴定时间间隔对麝香抗凝血酶活性测定的影响,并比较不同品种和产地麝香抗凝血酶活性的大小。结果显示,麝香供试品溶液浓度在一定范围内(0.01~0.02 g.mL-1),纤维蛋白原浓度为0.5%,凝血酶浓度为10 U.mL-1,滴定间隔时间为1 min,每次滴定2 μL时,麝香供试品溶液浓度与凝血酶消耗体积呈良好的线性关系(r=0.9914)。不同产地林麝麝香抗凝血酶活性效价为104.9±10.5 U.g -1,马麝麝香抗凝血酶活性效价为100.9±7.6 U.g -1,原麝(安徽)麝香抗凝血酶活性效价为97.6 U.g -1,人工麝香抗凝血酶活性效价为58.5 U.g -1。结果表明,抗凝血酶活性效价测定方法简便、快速、准确、可靠,可用于麝香质量生物评价;品种和产地对麝香抗凝血酶活性影响较大,在麝人工饲养时应注意品种和产地的选择;人工麝香和天然麝香的抗凝血酶活性差异较大,提示人工麝香处方配比可能有待进一步优化。

Quality evaluation of musk based on the biological potency of its antithrombin activity
Abstract:This study attempts to establish a method for the antithrombin activity bioassay of musk, explore the impact of species and producing areas on the antithrombin activity of musk, and provide scientific basis for its biological quality evaluation. Antithrombin activity of musk was analyzed by thrombin titration, and theSinfluence factors such asS musk solution concentration, fibrinogen concentration, thrombin concentration and titration interval were optimized to evaluate the effect of different species and producing areas on antithrombin activity of musk. As a result, There was a good linear relationship between musk solution concentration and thrombin consumption volume within the range of 0.01~0.02 g.mL-1 (r=0.9914) when fibrinogen concentration was 0.5%, the thrombin concentration was 10 U.mL-1, titration time interval was once every minute, each titration volume was 2 μL. The average antithrombin activity potency of Moschus berezovskii from different producing areas was 104.9±10.5 U.g -1, 100.9±7.6 U.g -1 for M. sifanicus from different producing areas, 97.6 U.g -1 for M. moschiferus from Anhui province, and 58.5 U.g -1 for artificial moschus. The results indicated that this antithrombin activity bioassay method could be applied to evaluate the antithrombin activity of musk quickly, conveniently, sensitively and exactly. It was also suggested that different species and producing areas had effects on the antithrombin activity of musk, so it is necessary to pay attention to species and producing areas in the process of musk domestication; meanwhile, the artificial musk and natural musk also showed great differences in antithrombin activity, suggesting that ratio in artificial musk prescription needs to be further optimized.
