中文摘要:本文采用整合药理学方法,预测中药复方越鞠丸作用的靶点、通路,探讨该方治疗抑郁症和胃肠道功能异常的 异病同治 分子机制。通过Human Phenotype Ontology数据库收集疾病靶标,用整合药理学平台进行药物靶标、GO功能、KEGG通路、核心靶标网络和中药-成分-关键药靶-通路网络的预测和分析,得到ATP1A1、KRAS、PRKAA1等关键靶点,富集到神经元凋亡过程、神经营养因子信号通路、5-羟色胺能突触、一氧化氮合酶活性的调节以及胆汁分泌、胃酸分泌等与抑郁症和胃肠道功能障碍相关的功能和通路,并推测5-羟色胺能突触和一氧化氮合酶活性的调节是抑郁症和胃肠道功能障碍的共同过程。本文基于整合药理学分析方法对越鞠丸有了整体性的认识,有助于阐明其对于抑郁症和胃肠道功能异常异病同治的作用机制。

A Study on Yuejuwan Pill for "Different Diseases with the Same Treatment" Underlying integrative pharmacology
Abstract:In this study, a integrative pharmacological method was adopted to predict the target genes and pathways of Yuejuwan Pill, and explore the molecular mechanism of depression and gastrointestinal dysfunction with the same treatment. First, disease targets of two diseases were collected from Human Phenotype Ontology database. Then, TCM-IP which is a integrative pharmacological platform were used to predict drug targets, GO function, KEGG pathway, core targets network of Yuejuwan and heterogeneous network of TCM – chemical components - key drug targets – pathway relationship. According to the integrative analysis, we found that ATP1A1, KRAS, and PRKAA1 were key targets; Neuron apoptotic process, Neurotrophin signaling pathway, Serotonergic synapse and Regulation of nitric-oxide synthase activity were key pathways which play important roles in molecular mechanism of depression and gastrointestinal dysfunction treated with Yuejuwan Pill. In conclusion, Speculated Serotonergic synapse and Regulation of nitric-oxide synthase activity maybe the common process of depression and gastrointestinal dysfunction.
